KO Validation: Overview
"KO validation" stands for "Knockout validation", which is a widely accepted method for determining the specificity and functional relevance of biological reagents—especially antibodies. The "KO validation grade" of a biological reagent is a measure of how well it has been validated using knockout models, and it provides confidence that the reagent specifically recognizes its intended target.
What Is KO Validation?
KO validation uses gene knockout cell lines or animal models, where a specific gene has been deliberately inactivated (knocked out). A reagent (usually an antibody) is considered KO validated if:
· It produces a positive signal in wild-type (WT) samples.
· It produces no signal or significantly reduced signal in knockout (KO) samples.
This approach provides direct evidence of specificity, because any signal in the KO sample likely indicates non-specific binding.
