Plasmapheresis experiment

Summary

Exudate may contain a large amount of plasma membrane proteins (also known as acidic glycoproteins Pr) whose isoelectric point is between PH 3-5 and can produce a white misty precipitate under acidic conditions. This experiment is derived from the Mudanjiang Medical College undergraduate 5-year clinical laboratory guide.

Principle

Exudate may contain a large amount of plasma membrane protein (also known as acidic glycoproteins Pr) whose isoelectric point is between PH 3-5, and may produce a white misty precipitate under acidic conditions.

Move

I. Experimental operations: 100ml of distilled water in a measuring cylinder, add 2-3 drops of ice acetic acid and mix. Then add one drop of the specimen to be tested in the measuring cylinder, and immediately observe under the black background, such as seeing a thick white cloudy precipitate falling rapidly, and no longer precipitate for Kivalta (+), such as the production of white turbidity is not obvious sinking slowly, and disappeared relatively quickly for Rivalta (-)

Second, the results of the experiment to determine:

clear does not show haze (-) Gradually show haze (Shi) After adding acid show haze (+) (-)

Gradually show fog (Shi)

Add acid show fog (+)

white book cloudy (+ + +)

white dense cloudy (+ + + +)

Caveat

1. Hemorrhagic plasma membrane fluid should be centrifuged and precipitated, and the supernatant should be used for the test.

2. Ascites of certain patients with cirrhosis of the liver, due to the high content of globular Pr, is cloudy and turbid, presenting a false-positive result.
