Overview
Fatty acid synthase (FAS, EC 2.3.1.85) is a key enzyme in fatty acid synthesis, catalyzing the formation of long-chain fatty acids from acetyl-CoA and malonyl-CoA. FAS is ubiquitously expressed in various tissues and cells, with abundant expression in mammalian liver, kidney, brain, lung, mammary gland, and adipose tissue.
FAS catalyzes the conversion of acetyl-CoA, malonyl-CoA, and NADPH into long-chain fatty acids and NADP⁺. Since NADPH exhibits an absorption peak at 340 nm, the activity of FAS can be determined by measuring the rate of decrease in absorbance at 340 nm.
