Overview
L-fucokinase/GDP-fucose pyrophos-phorylase (FKP, E.C. 2.7.1.52/2.7.7.30), derived from Bacteroides fragilis.This bifunctional enzyme catalyzes the phosphorylation of L-fucose to form Fuc-1-P, and further converts Fuc-1-P to GDP-Fuc, which serves as a fucosyl donor for glycosyltransferases. It acts as a tool enzyme for the synthesis of GDP-Fuc and its derivatives. This product is recombinantly expressed in Escherichia coli, featuring high purity and good compatibility, suitable for one-pot multi-enzyme systems.
Reaction:
Protocol for Use
Add substrates to the reaction mixture, supplemented with 5 mM MgCl₂. An appropriate buffer (e.g., Tris-HCl) may be added to adjust the pH to 7.5–8.5, followed by the addition of an appropriate amount of FKP (adjust according to reaction requirements).
Mix thoroughly and incubate the sample at 37°C for 30–60 minutes. The enzyme exhibits maximum activity in near-neutral pH buffers. The recommended reaction buffer is Tris-HCl (pH 8.0); other common biological buffers such as Tris or PBS are also applicable.
Note: For scaled-up reactions, adjust the incubation time or enzyme amount based on actual conditions. Gentle shaking may be used for mixing. Addition of inorganic pyrophosphatase can improve the conversion rate.
