There are various methods for staining mucous substances, such as AB-PAS staining, mucin HID-AB staining, and standard Alcian blue staining. Most of these methods rely on the principle that Alcian blue, a cationic dye, can bind to acidic groups-specifically, Alcian blue forms insoluble complexes with anionic groups (e.g., carboxyl groups and sulfate groups) present in tissues.
Mucin Staining Solution (Mild Methylation Method) belongs to the category of chemical modification and blocking methods. Its principle is to use an alcohol-containing alkaline solution to cleave the O-acetyl groups in enzyme-resistant sialic acid. The removal of O-acetyl groups and the formation of hydroxyl groups on the side chains of sialic acid restore the reactivity of sialic acid to PAS staining. When the pH value of Alcian staining solution is 2.5, the carboxyl groups in tissues undergo ionization and carry a negative charge, forming salt bonds with the cations in Alcian blue. This enables the staining of tissues containing carboxyl groups (e.g., proteoglycans/hyaluronic acid and epithelial acidic mucins), and the solution is mainly used to identify acidic groups in mucins. This reagent is for research use only and not intended for clinical diagnosis or other purposes.
Materials to Be Prepared by the User
1. Graded ethanol solutions, distilled water, deionized water, xylene or environmentally friendly dewaxing-clearing solution, mounting medium
2. Incubator, microscope
Operating Procedures (For Reference Only)
1. Dewax two positive control sections and two experimental sections to water.
2. Place one positive control section and one experimental section in Acidic Methanol Solution and incubate at 37℃ for 4 hours; incubate the remaining one positive control section and one experimental section in deionized water at 37℃ for 4 hours only. Rinse with running water for 5 minutes.
3. Stain with Alcian Staining Solution for 15-30 minutes. Rinse with running water.
4. Dehydrate with graded ethanol solutions, clear with xylene or dewaxing-clearing solution, and mount with neutral gum.
Staining Results
Untreated sulfated mucins, sulfated proteoglycans, sialomucins, and hyaluronic acid: Blue
Treated sections: Light blue
Note: Any residual color in the treated sections is due to the presence of sulfated mucins and/or sulfated proteoglycans.
Precautions
1. Incubation time exceeding 4 hours may cause hydrolysis of sulfate groups.
2. Positive control sections are required to verify the effectiveness of the methylation procedure.
3. For your safety and health, wear a lab coat and disposable gloves during operation.
