Proline (Pro) is a cyclic α-imino acid with a neutral charge and an isoelectric point of 6.30. Its solubility in water exceeds that of any other amino acid, with approximately 162g soluble in 100g of water at 25°C. It is hygroscopic, does not readily form crystals, and has a sweet taste. When heated with ninhydrin solution, proline produces a yellow compound. Upon incorporation into a peptide chain, it can undergo hydroxylation to form 4-hydroxyproline, a crucial component of animal collagen. Hydroxyproline is also present in various plant proteins, particularly those involved in cell wall formation. Under normal conditions, proline levels are relatively low; however, under stress conditions such as drought, heat, cold, freezing, or salinity, a significant accumulation of proline is often observed. This accumulation index is correlated with plant stress resistance. Proline has wide applications in clinical, biomaterial, and industrial fields. The detection principle of this kit is based on the reaction between free proline in a sulfosalicylic acid solution and acidic ninhydrin upon heating, which generates a stable red product. The absorbance at 520 nm is measured using a spectrophotometer, and within a certain concentration range, the color intensity is proportional to the proline concentration. This kit is primarily used for determining the free proline content in plant tissues and is intended for research purposes only. It is not suitable for clinical diagnosis or other applications.
