Safranin O is a cationic dye that binds to polyanions. Basophilic cartilage binds to the basic dye safranin O and stains red. It is often used in combination with fast green to visualize cartilage, based on the binding of the cationic dye to the anionic groups in mucopolysaccharides. Eosinophilic bone binds to the acidic dye fast green and stains green or blue, creating a sharp contrast with the red-stained cartilage, thus distinguishing cartilage tissue from bone tissue. It can also be used for the staining of plant specimens and mast cells.
