Overview

Saponification solution is primarily composed of ethanol and strong alkali. Saponification is a type of chemical modification and blocking method. Its principle involves using an alcohol-containing alkaline solution to cleave O-acetyl groups in enzyme-resistant sialic acids. The removal of O-acetyl groups and the formation of hydroxyl groups on the sialic acid side chains restore the reactivity of sialic acids to PAS. Saponification technology is also used to reverse methylation by cleaving the methyl ester bonds formed during the methylation process and reforming carboxyl groups. This reagent is intended for research purposes only and is not suitable for clinical diagnosis or other applications.

Procedure (for reference only)

Operate according to the specific experimental requirements. Typically, treat at room temperature for 30 minutes.

Precautions

  1. Positive control slides are required to verify the effectiveness of the methylation procedure.

  2. For your safety and health, please wear a lab coat and disposable gloves during operation.

  3. After opening, the reagent should be used as soon as possible to avoid affecting subsequent experimental results.
