Seed viability (often referred to simply as seed vigor) refers to the potential ability of a seed to germinate or the vitality of the embryo. The level of seed viability determines seed quality and practical value, and it influences the amount of seed required for sowing. A common method for determining seed viability is the germination test, where seeds are allowed to imbibe water and germinate under suitable conditions, and the percentage of germinated seeds out of the total tested seeds is counted within a specified number of days. TTC is a redox indicator with a standard oxidation-reduction potential of 80 mV. It forms a colorless solution when dissolved in water, and upon reduction, it produces red, water-insoluble triphenylformazan. This compound is relatively stable and not easily oxidized. Therefore, TTC is widely used as a hydrogen acceptor in enzymatic studies. The amount of TTC reduction reflects dehydrogenase activity, thereby serving as an indicator for assessing the viability of seeds from plants such as wheat, mung beans, rice, rapeseed, and peanuts. When the TTC staining solution penetrates the cells of the embryo, the embryo is stained red. When embryo viability decreases, respiration weakens significantly, dehydrogenase activity declines, and the color change in the embryo is less pronounced. When the embryo is non-viable, it remains unstained. Thus, the degree of staining indicates the level of seed viability.
