Overview

  Clinically, the integrity of the sperm membrane can be assessed to evaluate sperm vitality. When sperm are placed in a hypo-osmotic solution, water molecules enter the sperm through the membrane, causing an increase in sperm volume and swelling. This swelling is particularly evident in the tails of normal sperm. In contrast, sperm with impaired membranes or dead sperm typically do not swell.

  Sperm Vitality Detection Solution (HOS Method), also known as Sperm Hypo-osmotic Swelling Solution, is composed of sodium citrate, D-fructose, and other components. Utilizing the principle of hypo-osmotic swelling described above, it effectively analyzes sperm survival rates. This kit is intended for research use only and is not suitable for clinical diagnosis or other purposes.

Procedure

1. Take a clean small test tube and add 1 mL of Sperm Hypo-osmotic Swelling Solution. Pre-warm at 37°C for 5 minutes.

2. Add 0.1 mL of liquefied semen sample. Mix gently and incubate at 37°C for 30 to 60 minutes.

3. Observe under a phase-contrast microscope. Sperm exhibiting swelling are identified by changes in tail morphology and are considered viable.

4. Count 200 sperm and calculate the percentage of swollen sperm (viable sperm) among them.

  Reference Range: Within 30-60 minutes after ejaculation, over 70% of sperm should be motile. In the Hypo-osmotic Swelling Test, over 60% of sperm should exhibit tail swelling.

Precautions

  1. Sperm vitality should be assessed immediately after semen liquefaction. It is best to perform the test within 30 minutes, and under no circumstances should it exceed 1 hour after liquefaction. Delays may lead to sperm deactivation due to dehydration or temperature changes, resulting in inaccurate results.

  2. To avoid repeated freeze-thaw cycles, which may compromise the reagent's efficacy, aliquot the solution into 1 mL portions after the first thaw and store frozen.

  3. Use the reagent promptly after opening the vial to prevent potential effects on subsequent experimental results.
