Urobilinogen, also known as urochrome, is formed when aged red blood cells are broken down in the liver or spleen. Hemoglobin is converted into indirect bilirubin, which is then metabolized by intestinal bacteria into urobilinogen.
Detection Principle
Under acidic conditions, Ehrlich's reagent reacts with urobilinogen to produce a red-colored compound. This kit is highly sensitive and is primarily used for the qualitative detection of urobilinogen in human or animal urine. This kit is for research use only and is not suitable for clinical diagnosis or other purposes.

Operating Steps (For Reference Only)
1. (Optional) If the urine sample contains bilirubin, it should be removed. Take 4 mL of fresh urine into a test tube, add approximately 1 mL of Barium Chloride Solution, mix well, centrifuge at 3000 g for 5 minutes, discard the precipitate, and keep the supernatant for use.
2. Take 2 mL of fresh, bilirubin‑free urine, add 0.2 mL of Ehrlich's Reagent, let stand for 10 minutes, and observe the result.
Result Interpretation
Observation method: Observe from the top of the tube against a white background, looking directly to the bottom.

Note: Normal urine should give a positive (+) result.
Precautions
1. Urine must be fresh. Urobilinogen oxidizes to urobilin upon prolonged standing, leading to false‑negative results.
2. If the urine contains interfering substances such as porphobilinogen, add 2 mL of chloroform after adding Ehrlich's Reagent. Shake well and let stand. Urobilinogen will transfer to the chloroform layer, confirming a positive result.
3. If antibiotics have suppressed the intestinal flora, urobilinogen may be reduced or absent.
4. Please use the reagent as soon as possible after opening to avoid affecting subsequent experimental results.
5. For your safety and health, please wear a lab coat and disposable gloves during operation.
