Overview

  ADP-specific Glucokinase is an ADP-dependent glucokinase with EC number 2.7.1.147. Its gene is derived from Thermococcus litoralis. It can use ADP as the phosphate donor to catalyze the conversion of glucose to glucose-6-phosphate. This product is recombinantly expressed in E. coli.

Unit definition: One unit of enzyme activity is defined as the amount of enzyme required to consume 1.0 μmol of ADP per minute under conditions of 50°C and pH 7.5.

Usage Instructions

1. Add the substrate and 10 mM MgCl₂ to the reaction system; adjust the pH to 7.5 with appropriate buffer salts, then add a suitable amount of  ADP-specific Glucokinase (as needed for the reaction);

2. After mixing, incubate the sample at 50°C for 4 hours or overnight.

【Note】 When scaling up the reaction system, adjust the incubation time or enzyme dosage according to actual conditions; low-speed shaking can be used for mixing.

【Note】 Most common biological buffers (e.g., Tris or PBS) are applicable. Buffers outside this pH range (e.g., acetate buffer) may be usable, but the incubation time or enzyme dosage needs to be optimized based on actual conditions.
