Reticulocytes are immature red blood cells. Due to the residual basophilic substances such as ribosomes in the cytoplasm, blue-green dot, linear or reticular structures can be seen after staining with brilliant tar blue solution. The more dot structures of reticulocytes, the younger the cell is, and the more mature the cell is.
Operation process:
1 Sample handling
1.1 For paraffin sections: 1.1.1 Dewaxing with xylene, 2×5~10min;
1.1.2 100% ethanol hydration for 5min, 95% ethanol for 2min, 70% ethanol for 2min;
1.1.3 Rinse with PBS for 2min.
1.2 For frozen sections:
1.2.1 Rinse with PBS for 2min.
1.3 For cultured cells:
1.3.1 Fixed with 4% paraformaldehyde for more than 10 minutes;
1.3.2 Rinse with PBS for 2×2 min;
2 Brilliant tar Blue dyeing solution For the above treated samples, brilliant tar blue dyeing solution for 5-30min.
3 Dehydrate, transparent and seal. Direct differentiation and dehydration of anhydrous ethanol, 3~5min; Xylene transparent 3~5min, neutral gum seal.
Note:
1 Need to bring 4% paraformaldehyde, gradient ethanol, xylene, neutral gum or other sealing tablets. 2 When using this kit for the first time, it is recommended to take 1 to 2 samples for pre-test.
