Overview

Information

Buflomedil HCl Buflomedil HCl is a vasodilator used to treat claudication or the symptoms of peripheral arterial disease; A nonselective alpha adrenergic receptor inhibitor.
In vitro

Buflomedil increases intracellular levels of cAMP in human keratinocytes, either secondary to phosphodiesterase inhibition (PTX) or adenyl-cyclase stimulation (BC). Buflomedil diminishes TPA-induced TNF-alpha and IL-8 release from keratinocytes; in the case of IL-8 it is possible that this inhibition occur to transcriptional level. Buflomedil significantly inhibits epinephrine-induced aggregation at the micromolar level. Buflomedil results in a weak inhibition of ADP- and collagen-induced aggregation. Buflomedil inhibits granular secretion and the interaction of fibrinogen with its receptor on platelet. Buflomedil hypochloride improves the capillary perfusion in such related situations, evoking a possible effect upon neutrophils. Buflomedil decreases 100% of histamine related neutrophil adhesion (flow system) and partially inhibits adhesion after IL-1-4 hours (flow and stable systems). Buflomedil decreases the expression of P-selectin at the surface of endothelial cells and hypoxia induced neutrophil adhesion.

In vivo

Buflomedil (6 mg/kg) increases flap survival in nicotine treated rats. Buflomedil (3 mg/kg) significantly reduces both post-ischemic capillary perfusion failure and leukocyte adherence in venules. Buflomedil reduces the vasodilation evoked by phentolamine and potentiates its depressive effect on vasomotion frequency and amplitude in the hamster cheek pouch.
