The heparin/heparan sulfate disaccharide standard is an indispensable disaccharide reference standard for the composition analysis and structure identification of heparin and heparan sulfate disaccharides. We used heparin enzyme to degrade heparin and heparan sulfate with different sulfation modes, and combined with gel filtration and anion exchange HPLC, we prepared high-purity C4, 5 unsaturated disaccharide standards with various sulfation modes. Including: eight common unsaturated disaccharides after heparinase degradation of natural heparin/heparan sulfate: I-A (Δ UA-2S - [1 → 4] - GlcNAc-6S), II-A (Δ UA - [1 → 4] - GlcNAc-6S), III-A (Δ UA-2S - [1 → 4] - GlcNAc), IV-A (Δ UA - [1 → 4] - GlcNAc), I-S (Δ UA-2S - [1 → 4] - GlcNS-6S), II-S (Δ UA - [1 → 4] - GlcNS-6S), III-S (Δ UA-2S - [1 → 4] - GlcNS), and IV-S (Δ UA - [1 → 4] - GlcNS) 4] - GlcNS, as well as unsaturated disaccharides such as I-H (Δ UA-2S - [1 → 4] - GlcN-6S), II-H (Δ UA - [1 → 4] - GlcN-6S), III-H (Δ UA-2S - [1 → 4] - GlcN), and IV-H (Δ UA - [1 → 4] - GlcN), which are deacetylated products during the processing of heparin/heparan sulfate; 2-Aminobenzamide (2AB) fluorescent labeling standard for various heparin/heparan sulfate disaccharide standards mentioned above.
