Overview

RPMI 1640 medium was developed by Moore et al. in 1967 at the Roswell Park Memorial Institute (RPMI) in Buffalo, New York, USA. In this designation, "RPMI" refers to the series of media developed by the Institute, while "1640" denotes the specific product catalog number. This medium is a modified version of McCoy's 5A, utilizes a bicarbonate buffering system, and—unlike most mammalian cell culture media—was originally formulated to a pH of approximately 8.0. Initially designed specifically for lymphocyte culture, RPMI 1640 has now been widely adopted for the cultivation of various normal and cancer cells (particularly suspension cells) and remains one of the most extensively used basal media worldwide. This product is formulated without phenol red. Phenol red is commonly employed as a pH indicator (appearing yellow at low pH, purple at high pH, and red at pH 7.2–7.4, which is optimal for cell growth). However, because it may mimic estrogenic effects (influencing estrogen‑sensitive cells), interfere with flow cytometric analysis, and affect sodium‑potassium balance in serum‑free media, this formulation deliberately omits the component to accommodate a broader range of experimental requirements. The concentrations of major components in this product are as follows: D‑Glucose:  4000 mg/L L‑Glutamine:  600 mg/L Sodium Bicarbonate:  2000 mg/L The final pH of the product is 7.0–7.4. This product contains various amino acids, vitamins, inorganic salts, and other nutrients required for cell culture; however, it does not contain proteins, lipids, or any growth factors. Therefore, supplementation with serum or serum‑free additives is necessary during use to meet cellular growth requirements. Precautions 1. This product has been sterilized by filtration. Please strictly adhere to aseptic techniques during handling to prevent contamination. 2. To maintain optimal performance, avoid repeated freeze‑thaw cycles. 3. For research use only (or for further investigative purposes); not intended for diagnostic or therapeutic applications.
