Product Description
Super Green I (20× DMSO solution, PCR grade) (the effect is the same as Sybr Green I) Combining with dsDNA, the fluorescence signal can be enhanced 800-1000 times. In the PCR reaction system, an excessive amount of Super Green I fluorescent dye is added. After it is specifically incorporated into the DNA double-strand, the fluorescent signal is enhanced, while the fluorescence of the SUPER Green I dye molecule that is not incorporated in the chain remains unchanged, thereby ensuring the fluorescence signal The increase is completely synchronized with the increase of PCR products. Fluorescence can be measured in the annealing stage or the extension stage.
1. The influence of concentration on the result of fluorescence PCR
The concentration of Super Green I is a very critical factor to ensure the success of fluorescent quantitative PCR experiments. If the concentration of SUPER Green I is too low, the change in fluorescence signal will be reduced, which means that low-copy samples may not be detected. At high concentrations, the PCR reaction will be inhibited and the PCR reaction efficiency will be reduced. Therefore, when using SUPER Green I, the concentration should be optimized according to the actual situation. The final concentration of the reaction is between 1× and 0.2×.
2. The influence of Mg2+ concentration
Increasing the concentration of Mg2+ can reduce the inhibitory effect of SUPER Green I on the PCR reaction. We recommend that when using SUPER Green I for fluorescent PCR reactions, the Mg2+ concentration is 0.5-3 mM higher than that of ordinary PCR reactions without SUPER Green I.
