Preparation of media with dry powder

Summary

Dissolve all the dry powder in the package with appropriate amount of ultrapure water: put the container on the magnetic stirrer and add the dry powder while stirring. When all the ingredients are completely dissolved, the medium should be filtered immediately and not left to stand, otherwise precipitation or microbial contamination may occur. When the final ingredient (e.g., glutamine, NaHCO3, or serum) has been added, it is best to adjust the pH .

Principle

Dissolve all the dry powder in the package with appropriate amount of ultrapure water: put the container on the magnetic stirrer and add the dry powder while stirring. When all the ingredients are completely dissolved, the medium should be filtered immediately and not left to stand, otherwise precipitation or microbial contamination may occur. When the final ingredient (e.g., glutamine, NaHCO3, or serum) has been added, it is best to adjust the pH .

Move

1. Add an appropriate amount of ultrapure water to the container.

2. Place the magnetic stirrer.

3. Place the container on the magnetic stirrer and set the speed to 200 r/min.

4. Open the package of dry powder and add the dry powder to the container slowly, stirring as you add.

5. Stir until the dry powder is completely dissolved.

6. Test the pH and conductivity of the sample and make a record.

(a) The pH should not change by more than 0.1 unit for a predetermined value for a given medium.

(b) Conductivity should not change by more than 2 % for a predetermined value in the specified medium.

(c) Discard the sample for testing and do not add back to the stock solution.

7. Sterilize by filtration.

8. cap and seal the bottle. ,

9. Store at 4°C.

10. Add serum and glutamine, if desired, and adjust pH to 7.4 before use.
