Overview

  Elastic fibers are mainly distributed in the arterial wall, alveolar wall, and skin of the human body. They appear yellow when fresh and possess strong refraction. Common staining methods for elastic fibers include Gomori Aldehyde Fuchsin method, Resorcin Fuchsin method, Orcein method, Victoria Blue method, Iron Iodine Hematoxylin method, etc. Elastic fiber staining can demonstrate changes of elastic fibers in skin tissue, such as elastofibroma, annular granuloma of the skin, and scleroderma; it can reveal and evaluate endocardial and arterial lesions, and observe whether certain lesions are accompanied by hyperplasia or destruction of elastic fibers. It can also be used to identify tumor components such as elastofibroma, in which elastic fiber globules can be clearly seen after elastic fiber staining.

  Lichen Red Elastic Fiber Staining Solution is easy to operate and highly specific, but the staining effect may vary with different batches of raw materials, and sometimes it may stain lipofuscin in liver cells. This reagent is for research use only. Not for clinical diagnosis or other purposes.

Immediately before use, mix equal volumes of Solution A and Solution B to prepare Shikata Acid Solution. Prepare fresh and use immediately.

Materials Required:

10% neutral formalin fixative, distilled water, graded ethanol, xylene or eco-friendly dewaxing and clearing agent, neutral balsam

Protocol (For Reference Only):

1. Fix tissues in 10% neutral formalin fixative, then process for routine dehydration and embedding.

2. Cut sections at 4–5 μm thickness. Deparaffinize routinely with xylene or dewaxing agent and rehydrate to water.

3. Oxidize in Shikata Acid Solution for 5 min, then rinse briefly in water.

4. Bleach in Shikata Washing Solution for 2 min to remove color from the acid solution, then rinse in running water for 2 min.

5. Rinse briefly in 70% ethanol.

6. Stain covered in Orcein Staining Solution for 5 h or longer.

7. Dehydrate routinely, clear in xylene or dewaxing and clearing agent, and mount with neutral balsam.

Staining Results:

Elastic fibers: Brownish red to dark brown

Background: Grayish brown or pale pink to varying degrees

Precautions: 

1. Shikata Acid Solution must be prepared fresh immediately before use. Prolonged standing after mixing will reduce its oxidizing activity.

2. Acid Orcein Staining Solution should be returned to room temperature before use. The staining solution can be reused several times.

3. After 5 h of staining, sections may be rinsed briefly in 70% ethanol, washed in distilled water, and examined microscopically. If staining is faint, sections may be rinsed in 70% ethanol and returned to Acid Orcein Staining Solution; overnight staining is acceptable if necessary.

4. Lipofuscin and HBsAg-positive substances may also stain positively with orcein. They can be distinguished as follows:

Lipofuscin: intensely stained, small granules of uniform size in the cytoplasm

HBsAg: weakly stained, fine granular pattern

Elastic fibers: intensely stained, non-granular

5. Use reagents promptly after opening to avoid affecting experimental results.

6. For your safety and health, wear a lab coat and disposable gloves during operation.
