Animal model of neonatal bilirubin encephalopathy

Summary

Bilirubin encephalopathy is a serious complication of neonatal yellow pox with high mortality, readmission and sequelae rates, and the survivors are often left with serious neurological sequelae, such as hearing impairment, learning disabilities, mental retardation, cerebral condition and even "tetralogy of Fallot". The animal model of neonatal bilirubin encephalopathy is usually made by injecting bilirubin solution into the cerebellar medulla oblongata pool of newborn rats and guinea pigs.

Principle

Injection via the medullary pool of the cerebellum causes bilirubin to enter the cerebrospinal fluid circulation directly and to be deposited in the central nervous system, leading to brain damage, known as bilirubin encephalopathy.

Materials and Instruments

Equipment:
Syringes, needles.
Reagents:
① Bilirubin;
② Hydrochloric acid;
③ Sodium hydroxide
④ Ether;
⑤ Alcohol.

Move

The basic process of establishing an animal model of neonatal bilirubin encephalopathy can be divided into the following steps:


A. 5 mg of crystalline bilirubin was dissolved in 50 μl of 0.5 mol/L sodium hydroxide, 450 μl of distilled water was added, and the pH was adjusted to 8.5 with 0.5 mol/L hydrochloric acid.


B. Pregnant SD rats were delivered at full term, and the mothers were fed until the 7th day. After weighing the newborn rats, they were anesthetized by inhalation of ether and placed in prone position. The back of the neck was sterilized and arched, and a syringe was used to insert the needle from the midpoint between the external occipital ridge and the highest point of the cervical vertebrae, and then the needle was inserted in the direction of the midpoint of the line connecting the two ears.


C, 2 μ/g of biliverdin was injected through the medullary pool of the cerebellum, and the neurobehavioral changes were observed.
