Summary

Sulfosalicylic acid is an alkaloidal reagent that can precipitate proteins, but the ability to precipitate white Pr is stronger than ball Pr. After adding Na2SO4, the same concentration of ALB and ball Pr were turbid and tended to converge, and quantitatively determined by comparing the concentration with the standard Pr. Principle

Sulfosalicylic acid is an alkaloid reagent, can precipitate proteins, but the ability to precipitate white Pr is stronger than the ball Pr, after adding Na2SO4, the same concentration of ALB and ball Pr was turbid, convergence, and quantitative determination in comparison with the standard Pr concentration.

Move

1. Take 4 test tubes, labeled as specimen tube, specimen blank tube, standard tube, reagent blank tube, add 0.5 ml of csf to be measured to the specimen tube and the specimen blank tube, add 0.5 ml of Pr solution to the standard tube, and 0.5 ml of saline to the blank tube.

2. Add 45 ml of SS-S reagent to each of the specimen and the reagent blank tubes, and mix thoroughly. Add 45 ml of saline to the specimen blank tube.

3. Allow to stand for 10 minutes and then shake well to measure the colorimetry at 520 nm, and adjust the absorbance zero point with saline. Determine the absorbance of the specimen blank tube VO to reagent blank tube to adjust the zero, determine the degree of the specimen tube (s) and specimen tube (V), according to the following calculations:

csf protein concentration, (mg / d) = (V - VO) X 24 urine volume/s.

Caveat

l. The cdf should be centrifuged first if there are a lot of cells or turbidity, and clear and transparent csf should be taken.

2. Before measurement should be done csf qualitative (+ + +) above. The result should be diluted 5-20 times with saline x the number of dilutions.

3. Temperature, time and reagent addition can affect the results, so it is not appropriate to use the standard curve method, it is advisable to bring the standard tube each time the same operation.
