Experiments on the action of drugs on isolated intestinal tubes

Summary

The purpose of this experiment is to master the experimental conditions of isolated intestinal smooth muscle organ and the effects of drugs on isolated intestinal smooth muscle; through the students' own design of the drug administration sequence, they will master the basic method of designing drug interactions, and at the same time observe whether the effects of barium chloride have any relationship with the receptor, so as to improve the ability of experimental design, and to understand the use of the equilibrium recorder.

Move

1. Each group discusses the design of the dosing sequence for the given drug, based on its action; afterward, the scheme is written on the board. Class discussion is held under the guidance of the teacher. Finally, the groups work out the best solution.

2. Preparation of the experimental setup This setup consists of thermostat, gas supply, liquid supply and discharge, and tracing device. The thermostat is completed by the Super Waki Thermal Bath, the gas supply is composed of an air pump and a hose, the liquid supply and discharge is composed of a water bath tube, which is connected with a hose and hose clamps at one end of the tube, and there is also a side tube at the top end of the tube that is connected with the hanging flask containing the Krebs liquid. The tracing device consists of a tension transducer and a bench-top automatic balancing instrument. The bench-top automatic balancing recorder is set to a range of 2 mv, and the tracing pen rises by 2 large frames (5 cm) by adding l g of weight.

3. Preparation of intestinal specimens

Take a healthy mouse, weighing about 20-30 g, dislocate the cervical vertebrae and make it die rapidly, immediately dissect the abdomen, and the peristaltic intestinal tube can be seen, cut several sections of 1-2 cm long small intestines, and put them in cold Krebs liquid for spare.

4. Installation of specimens hand washed, finger end stained with nutrient solution, take a section of small intestinal specimen, one end of the hook with a small hook with a line, and make the end of the line from the lower mouth of the bath tube hanging out, with a cork plug, and the other end of the hook in the line and the tension transducer connected to the hook, the hook should be through the intestinal wall symmetry of the sides of the small hook, adjusting the length of the line or the elevation of the lever position of the intestinal tube of the pre-added tension for the 0.5 g, that is, the desktop equilibrium recorder on the tracing pencil rise 1 If the temperature, air supply and nutrient solution are suitable and the intestinal segment is not dead, the intestinal tube will start to peristalsis a moment later, and a normal curve will be traced after the peristalsis is more constant.

5. 0.1 ml per dose. If the effect is not obvious, another 0.1 ml can be added.

6. Analyze and discuss the results of the experiment and write a report.

Caveat

1. the experiment should be performed under suitable conditions and the gas supply should be continuous.

2. intestinal tube specimens should be handled gently to maintain specimen activity.

3. syringes should not be mixed and drugs should be added directly to the liquid level, not reaching below it.

4. use the tension transducer and the benchtop auto-equilibrium recorder with care.

5. Acetylcholine is easily hydrolyzed, so add acetylcholine and wait until its effect is obvious, then add atropine to observe their interaction.
