Principle

Heterogeneous staining of particles: Neisser's staining method

Neisser staining method: Take the culture of Corynebacterium diphtheriae cultured in Lüscher's serum slant and mix the plate with physiological saline. Add the mixture of the first and second solution of Neisser's stain to the flame-fixed specimen, stain for 1-3 minutes, wash with water, and then stain with the third solution for half a minute, pour off the stain, and then dry for microscopic examination. The body of Corynebacterium diphtheriae was yellow-brown, and the heterochromatic particles were blue-black.
