Principle

Performing perfusion in experimental animals is necessary to obtain good morphological observations as well as to preserve the brain, kidneys, heart and many other organs.

Move

1. One syringe was filled with 1×PBS and the other with 4% PFA fixative (4°C) and set aside for use.

2. Mice were executed with CO2 gas in a bag. Immediately after stopping inhalation and exhalation, flatten the mice dorsally, carefully dissect the thorax to prevent excessive bleeding, carefully and quickly incise the ribs, cut off the diaphragm, and expose the heart.

3. Carefully insert a syringe of 1×PBS into the left ventricle and dissect the right ventricle to drain the fluid. Slowly but continuously instill 1 x PBS into the heart (if perfusion is normal, blood-rich organs such as the liver, spleen, and kidneys will turn grayish-white).


4. When most of the blood has drained out, the syringe of 1×PBS is removed and a syringe with 4% PFA fixative is inserted into the left ventricle at the same entry point, and the mouse is slowly perfused with 20 ml of fixative.

5. After perfusion, the organs or tissues were dissected out and placed in labeled glass vials containing 4% PFA fixative and stored on ice.

Caveat

For untrained experimenters, perfusion may fail the first time, so it is recommended to first practice with some control experimental animals to familiarize with this procedure.
