Preparation of pH standard color scale

Summary

Most cell lines grow best at pH 7.4, with little variation between cell lines; however, some normal fibroblast cell lines grow best at pH 7.4 to 7.7. However, some normal fibroblast lines grow best at pH 7.4 to 7.7, and some transformed cells may grow better at pH 7.O to 7.4 [Eagle 1973]. It has been reported that epithelial cells can be maintained at pHS.5 [Eisinger et al., 1979], but this criterion is not universally accepted. For specific conditions, the appropriate pH can be determined by simple growth assays, inoculation rate analysis, or specific function analysis, which may be more favorable for the growth of specific cells.

Principle

Phenol red is a commonly used indicator. At pH 7.4, it is red; at pH 7.0, it becomes orange; at pH 6.5, it is yellow; below pH 6.5, it is lemon yellow; at pH 7.6, it is peach; and at pH 7.8, it is purplish. Because of the highly subjective nature of color judgments, it is useful to make a standard color scale: it can be prepared from bacteriostatically inactivated balanced salt solution (BSS) and phenol red at conventional concentrations in identical bottles with the same amount of space remaining at the upper end, and it can generally be used in the preparation of culture media. The following scheme is based on the use of 25. resistant culture bottles as an example.

Hank's Balanced Salt Solution (HBSS), or Eagle's MEM 10x Concentrate, without carbonate or staphylin. Contains 20 mmol/L HEPES 10 ml.

Ultra Pure Water (UPw) 90ml

1mol/L NaOH 10ml

Procedure

1. Prepare H-cold vapor or medium, pH 6.5 or slightly below 6.5, and dispense into 7 labeled containers.

2. Equilibrate in air for 30min.

3. Adjust the liquid in each container with lmol/L NaOH H to pH 6.5, 6.8, 7.0, 7.2, 7.4, 7.6, 7.5, respectively, and test on the side of a pH meter.

4. Take each kind of HBSS, filter and sterilize, collect 5 ml of HBSS into labeled culture bottles, and rinse the filters with 5 ml of HBSS before collection.

5. Close the culture bottles.
