Toxic reactions to streptomycin and antidote experiments

Summary

Streptomycin is an aminoglycoside antibiotic, and its acute toxic reaction is neuromuscular blockage, with weakness of limbs and even respiratory depression. In this experiment, an overdose of streptomycin was injected to produce acute toxicity in mice, in order to observe the protective effect of calcium chloride on anti-streptomycin poisoned mice.

Principle

Streptomycin is an aminoglycoside antibiotic, and its acute toxic reaction is neuromuscular blockage, with weakness of limbs and even respiratory depression. In this experiment, the acute toxicity of streptomycin was produced in mice by injecting excessive amount of streptomycin, and the protective effect of calcium chloride against streptomycin poisoning in mice was observed.

I. Methodology

Two rabbits were taken, labeled, weighed, and the animals were observed for respiration, reflexes and muscle tone of the limbs. Rabbit A was injected 2.4 ml∕Kg of streptomycin sulfate solution intramuscularly, and its response was observed. When respiratory paralysis occurred, a rubber catheter connected to the air outlet of the artificial respirator was inserted into one side of the rabbit's nostril, and artificial respiration was given continuously to observe whether it could recover. Rabbit B was injected 2.4 ml∕Kg of streptomycin sulfate solution intramuscularly, and after respiratory paralysis, artificial respiration was also given through the nasal catheter, and 1.6 ml∕Kg of calcium chloride solution and 0.3 ml∕Kg of neostigmine methylsulfate were injected intravenously at the ear margin, and whether the rabbit could recover was observed.
II. Results

Caveat

After streptomycin is given intramuscularly, the reaction usually occurs in 30-60 minutes and progressively worsens. Calcium chloride solution should be pushed slowly to avoid hypercalcemic convulsions.
