Emotional State Screening Experiment
Emotional State Screening Experiment
Currently, there are four main approaches to mood state screening experiments: the opening experiment, the light and dark box test, the elevated cross maze, and the Porsolt swim test.
Principle
The basic principle of the open-field experiment is to detect mice's anxiety and exploratory behavior towards the outside world by recording the time they spend in the edge zone of the open-field environment and the time they spend in the central zone (anxiety-producing zone).

Materials and Instruments
Equipment: Move The basic procedure for the opening experiment can be divided into the following steps: Caveat 1. except for cage change, mice should not be subjected to any manipulation, such as scratching, prior to testing. 2. If ear marking is required, it should be done after this test. For more product details, please visit Aladdin Scientific website.
① Opening space.
② Video detection equipment.
③ Analyzing software.
A Transfer the mice to the assay room and keep them undisturbed for at least 30 minutes prior to assaying.
B Ensure that the light conditions are as required and that the apparatus is functioning properly.
(1) If using an infrared ray system, make sure that the rays are undisturbed before the mice are placed, and if detecting standing, make sure that the height of the ray port for standing is appropriate.
(2) If a video tracking system is used, debug the detection parameters to correctly distinguish between the mouse's fur color and background color.
(3) For post-test analysis, each open-field space was divided into a central zone and a marginal zone, with the marginal zone 8 cm wide from the wall and the central zone accounting for 40% of the total area.
C Start the test 1 hour after the light is turned on during the normal daily photoperiod, and the time of the test needs to be kept consistent every day.
D Clean each open-field zone and dry it.
E Place the mice into the open-field space one by one in a direction where the midpoint of the marginal zone is facing the wall on one side, and allow them to explore the open-field space freely. The experimenter should be out of sight of the mice. If a video tracking system is used to detect more than one mouse in parallel in adjacent spaces, it is important to ensure that data are recorded as soon as each mouse is placed to ensure consistency of the raw data. Male and female mice should be tested in separate batches, preferably alternating between males and females.
F After 20 minutes, remove the mice and return them to their original cages.
G Remove feces from the open field area and wipe all areas clean and dry, and move on to the next batch of mice.
H Calculate the activity of the mice in the different areas at 5-minute intervals by analyzing the recorded data.
