In vivo staining experiments of the vesicular lineage

Summary

In animal cells, all vesicles and vesicles surrounded by membranes except mitochondria belong to the vesicular lineage, including Golgi apparatus, lysosomes, microsomes, digestive vesicles, autophagic vesicles, remnants, cytosolic vesicles, and phagocytosis vesicles, all of which are surrounded by a unitary membrane.

Principle

Neutral red is a special living stain for the vesicular lineage, staining only the vesicular lineage red; the cytoplasm and nucleus are not stained when the cells are in the living state; neutral red staining may be related to proteins in the vesicles.

I. Live staining and observation of the vesicular lineage of chondrocytes in the sternal raphe of the toad (Bufo vulgaris)
1. Methodology
(1) Take a toad, destroy the brain and spinal cord, and cut open the thorax.

(2) A small piece of the thinnest part of the cartilage of the sternal raphe is taken and placed on a carrier sheet.

(3) Put two drops of 1/3000 neutral red staining solution, stain for 8~10 minutes and remove the stain with absorbent paper.

(4) Add one drop of Ringer's solution, cover the slide and aspirate the excess Ringer's solution.
2. Results: Under the microscope, chondrocytes were seen to be oval in shape, with many nuclei stained rose-red around the nucleus. Small vesicles of different sizes, i.e. chondrocyte vesicle lineage.
II. Electron microscopic photographs of chondrocytes in the tibial epiphysis of rats
1. Animal cell vesicles are so small that even when stained in vivo they can only be seen as a small dot. In order to further visualize the structure of the vesicles it is necessary to look at electron micrographs of chondrocytes.
2. Electron micrograph of rat tibial epiphyseal cells, with clear nucleus and nucleolus, abundant rough endoplasmic reticulum in the cytoplasm, well-developed vesicular lineage, vesicles surrounded by a single membrane, and vesicles released from the periphery of the cell.
