Operation method
Inhibition of respiration by niclosamide against dulcolax
Principle
Central stimulants increase the functional activity of the central nervous system. Among them, nikethamide mainly excites the respiratory center of the medulla oblongata (also known as respiratory excitatory drugs). When the respiratory center is inhibited, the effect of this drug is obvious, can make the respiration deepen and accelerate, and can improve the respiratory center's sensitivity to CO2.Diazepam is a central inhibitory drug, has the effects of anxiolytic, sedative-hypnotic, myorelaxation, anticonvulsant and antiepileptic, and so on. Its anticonvulsant by inhibiting the spread of abnormal discharges in the cerebral cortex, thalamus, limbic system, may be related to the promotion of a variety of synaptic transmission functions realized by g-aminobutyric acid (GABA).
Materials and Instruments
dolantin nikethamide dicaine hydrochloride diazepam Move 1. Experimental system connection and parameter setting Common Problems 1. Record a period of normal respiratory curve. The respiratory depression of rabbits was observed by intravenous injection of 5% Dolantin solution 50-100 mg/kg (1-2 ml/kg) from the ear margin of rabbits. Rabbit urinary catheter respiratory transducer microcomputer biosignal acquisition and processing system
(1) Fix the respiratory transducer on the iron pillar, and connect the output line of respiratory transducer to the 1st channel of microcomputer biological signal processing system (other channels are also available).
(2) Microcomputer biological signal processing system parameter settings
① RM6240 system: Click "Experiment" menu, select "Breathing" or "Breathing Record" in "Customized Experiment" menu. "The system enters the signal recording state of the experiment. The system enters the signal recording state of the experiment. Instrument parameters: channel time constant is DC, filter frequency is 30Hz, sensitivity is 10cmH2O (or 12.5ml), sampling frequency is 800Hz, scanning speed is 1s/div.
② PcLab and MedLab system: Click the "Experiment" menu, select "Common Physiology Experiments" or "File" menu "Open". Configuration" in the "Respiration Recording" item. The system enters the signal recording state of the experiment. Instrument parameters: channel amplification 1000, DC coupling (lower limit frequency DC), upper limit frequency 40Hz, sampling interval 1ms;
2. Animal Preparation
The rabbit was weighed and fixed in the rabbit fixation box. 1~2 drops of 1% dicaine solution was inserted into one side of the rabbit's nostril, then the head end of the catheter was coated with liquid paraffin and slowly inserted into the nostril. The other end of the catheter was connected to the respiratory transducer, and the cannula was inserted into the end of the nostril by rotating and adjusting the depth and angle of its insertion so that the respiratory curve of the rabbit had an appropriate amplitude.
2. When the respiratory depression is obvious, immediately inject 25% Nikethamide solution 0.4ml/kg intravenously from the ear margin of the rabbit, and observe and record the respiratory changes.
