Immunohistochemistry Washing Solution (PBST), also known as Immunohistochemistry Rinsing Buffer, is applicable for washing after the incubation of primary or secondary antibodies during immunohistochemical staining. Proper washing can reduce background interference and enhance the signal-to-noise ratio.
Immunohistochemistry Washing Solution (10×PBST) is formulated with PBS, an appropriate amount of Tween 20 and preservative PC300, which can permeabilize cell membranes. Cell samples treated with this product allow for the detection of intracellular target proteins, including those in the cytoplasm and nucleus. Based on the dosage of 10 ml PBST required for washing per sample group, one package of Immunohistochemistry Washing Solution (PBST) can be used for 50 washes. This reagent is for research use only (RUO) and not intended for clinical diagnosis or any other applications.
Operating Procedures (For Reference Only)
1. Take an appropriate amount of this product and add 9 volumes of deionized water to prepare the working solution of Immunohistochemistry Washing Solution (PBST).
2. After the incubation of primary or secondary antibodies, add an appropriate amount of Immunohistochemistry Washing Solution (PBST) to completely cover the samples.
3. Gently shake and rinse on a shaker for 5 minutes, then change the solution and repeat the rinsing twice, 5 minutes each time.
Precautions
1. If shaking is likely to cause sample detachment, the samples can be soaked in the washing solution without shaking; in this case, extend the rinsing time to 5-8 minutes and increase the rinsing frequency by 1-2 times. If the background interference remains high, the rinsing time and frequency can be further increased.
2. Use the product as soon as possible after opening, as the efficacy of the solution may decrease upon prolonged storage.
3. Generally, extending the washing time or increasing the washing frequency will not exert a negative effect on the results of immunostaining.
4. Use the reagent immediately after opening to prevent adverse impacts on subsequent experimental results.
