2. Place the container on the magnetic stirrer and set the speed to about 20 r/min.
3 . Open the package of D-PBS dry powder or take 10 tablets and slowly add them into the container while stirring.
4. Keep stirring until the dry powder is completely dissolved.
5. Test the pH and conductivity of the sample and record.
(a) The pH should not vary by more than 0.1 unit (pH may vary from formulation to formulation).
(b) Conductivity should not vary more than 5% of 15 μS/cm. Optionally, osmolality may be measured (or both) and should show similar conductivity between batches.
(c) Discard the sample for measurement, taking care not to add it back to the stock solution.
6. Dispense the liquid from the container into graduated bottles.
7 . Cap the bottles and seal them. It is useful to wrap the lid in paper with elastic adhesive and mark the paper by writing the date of the date and the name of the contents of the bottle, which also prevents the label from being removed from the bottle.
8 . Apply a small piece of autoclave tape or sterile indicator label and write the date.
9. Place the sealed bottle in an autoclave at 121°C, 100 kPa (1 bar, 15 lb/in2 ) for 20 min.
10. Store solution at room temperature.
