Prepare storage concentrates and freeze for preservation. Thaw, mix, aseptically filter and store when needed.

Materials and Instruments

1. Thaw the solution to ensure that all solutes are dissolved.

2 . Mix the ingredients in the correct proportions:

(a) Amino acid concentrate 100 ml

(b) Tyrosine and tryptophan 200 ml

(c) Vitamins 10 ml

(d) Glucose 100 ml

3. Mix and sterilize by filtration.

4. Store frozen.

5. For use, mix 41 ml of concentrate with 959 ml of 1X BSS.

6. Adjust pH to 7.4.

7. Store at 4°C.

8. Add human serum or other supplements, e.g. hormones, growth factors, lipids, before using the medium. If metals are used as trace elements, it is also advisable to add them before using the medium. This is because they will form a precipitate if the concentrate contains phosphates.
