Assay method for activity
1. Principle


The resulting Quinoneiminedye can be detected at 555nm.

2. Definition of enzyme activity
Unit enzyme activity is defined as the amount of enzyme required to catalyze the production of 1 μmol H2O2 per minute under the following conditions.

3. Reagent preparation
Reagent I: 0.2M Tris-HCl buffer, pH8.0.
Reagent II: 1.0M sarcosine solution (8.9g sarcosine dissolved in 100mLUP water).
Reagent III: 1kU/mL POD.
Reagent IV: 50 mm4-AA solution (1.016g4-AA dissolved in 100mLUP water).
Reagent V: 50mM TOOS solution (1.657gTOOS dissolved in 100mLUP water).
Enzyme diluent: 20mM Tris-HCl, pH8.0, 2mM EDTA.
Prepare the reaction mixture as follows:

Reagent I   5mL
Reagent II 10mL
Reagent III 0.25mL
Reagent IV 1.5mL
Reagent V 1.5mL double steaming water 31.75mL
A total of 50mL can be used for 50 reactions.

4. Operation procedure
1. Add 1 ml reaction mixture to 1mL colorimetric dish.
2.37°C temperature bath for 5min.
3. Add 20μL of enzyme liquid to the reaction mixture and mix well.
4.37°C reaction, detection of sample absorbance change within 1min at 555nm (∆As)
* Replace enzyme liquid with enzyme diluent, other steps are the same, the absorbance of the resulting solution is blank absorbance (∆Ab)
