Papanicolaou staining is commonly used for routine cytological staining. Papanicolaou Stain was initially used only to assess estrogen levels in vaginal epithelium and detect pathogens such as Candida and Trichomonas in the genital tract. After staining, the cytoplasm should be transparent and distinct, and nuclear chromatin should be easily identifiable. Current modified Papanicolaou staining solutions contain various ions and exhibit polychromatic staining effects. Stained cytoplasm is bright and transparent, with clear structures of the nuclear membrane, nucleolus and chromatin. Papanicolaou Stain is applied to exfoliated cell specimens, staining cell nuclei blue or black, and cytoplasm of keratinized squamous cells pink or orange‑red.
EA36 Staining Solution is mainly composed of light green, eosin, phosphotungstic acid and other components, and contains no acetic acid. It is used as a cytoplasmic stain. When used in combination with Orange G6 and EA36 or EA50, it can stain the cytoplasm bright green, blue and pink. EA36 Stain is more suitable than EA50 Stain for gynecological cytological smears, such as screening for cervical cancer and precancerous lesions.
