Ethanol serves as a critical analyte, and its precise detection is essential across multiple fields. In the brewing industry, ethanol content is a core indicator for evaluating the quality of alcoholic beverages, directly impacting product quality control and consumer health. In the legal and traffic safety sectors, blood ethanol concentration provides the legal basis for defining drunk driving and impaired driving. Furthermore, in postharvest physiology of fruits and vegetables, ethanol levels serve as an important parameter for monitoring storage quality and assessing the effectiveness of preservation treatments (for instance, appropriate ethanol treatments can delay senescence and inhibit disease). Achieving simple and rapid measurement of ethanol has practical significance for extending product shelf life. The detection typically employs an efficient enzyme-coupled colorimetric method: ethanol is oxidized to acetaldehyde under the catalysis of alcohol dehydrogenase (ADH), while simultaneously reducing oxidized nicotinamide adenine dinucleotide (NAD⁺) to NADH. In the presence of the electron mediator 1-methoxy-5-methylphenazinium methyl sulfate (1-mPMS), the generated NADH further reduces the water-soluble tetrazolium salt WST-8, producing an orange-yellow formazan product. This product exhibits a characteristic absorption peak at a wavelength of 450 nm. By measuring the rate of increase in absorbance at this wavelength, the ethanol content in the sample can be accurately calculated.
