Summary
Enzyme-linked immunospot test (ELISPOT) is based on the principle that cells that secrete certain protein molecules (e.g. cytokines) are able to secrete a relatively high concentration of protein molecules into their surroundings in a short period of time. basic program Material
Coating buffer
Wash buffer: PBS solution with 0.25% (v/v) Tween 20
Blocking buffer: PBS solution with 5 % (m/V) BSA or FCS
VRPMMO complete medium (or other suitable medium)
Cytokine-secreting cells (mouse or human)
Mitogens, antigens, or other stimuli that promote cytokine secretion
Cytokine-specific secondary antibodies with markers
