Experiments on the preparation of encapsulated tissue culture plates

Summary

This experiment describes the preparation of cutin A-encapsulated culture plates for growing CEF monolayers for in vivo immunostaining. Source Compact Molecular Biology Laboratory Guide (5th Edition)

Operation method

basic program

Materials and Instruments

Knife Bean Protein A
Phosphate buffer solution (PBS)
6-well 35 mm Tissue Culture Plates Sterile plastic bags (for preserving the coverslips)

Move

1. Add 12 mg of Knife Protein A to 120 ml of sterile PBS to a final concentration of 100 μg/ml.

2. Add 1 ml of PBS/Knife Protein A mixture to each well of the tissue culture plate and incubate at room temperature for 1 h. 3. Pipette each well with 2 ml of PBS.

3. Aspirate the liquid from each well and rinse with 2 ml of PBS. 4.

4. Aspirate the liquid and dry the plates in a biosafety cabinet (to maintain sterility).

5. Store the culture plate in a plastic bag (to maintain sterility) at room temperature (can be stored for several months).
