Handling experiments on pre-coated slides

Summary

Slides coated with polylysine (a Thermo product) with durable bioadhesion for attaching frozen and paraffin-embedded sections that can be subsequently centrifuged and subjected to cytological blotting assays.

Move

I. Polylysine-encapsulated planted slides1. Wash the slides (with slide holders and glass staining trays) as for gelatin-soaked treatment of slides.

2. Prepare a sufficient amount of 500 μg/ml poly-L-lysine aqueous solution.

3. Dip the slides one by one in this solution.

4. Dry in air and store at 4°C. Use within one week (plastic slide boxes can be used as dip boxes).II. Gelatin-impregnated treatment for loading slides1. Wash the slide (25 mm x 75 mm) for a few minutes in water with detergent, any good quality detergent can be used.2. Soak the slides in water for 30 min, during which time 1 x gelatin soaking solution is prepared.3. Place the slide in a slide holder and immerse it in a glass staining dish containing 1× gelatin soaking solution (4°C) for 2 min. The soaking process must prevent air bubbles from forming on the slide.
4. Remove the slide holder from the soaking solution, stand the slide on its side to allow the soaking solution to drip off, and dry overnight before use. Gelatin-soaked slides are stable at room temperature for several weeks.
