hemolytic reaction assay

Summary

Immune serum meets with its corresponding antigenic cells, blood cells, bacteria and their tissue cells, and a cytolytic reaction can occur with the participation of complement. Depending on the type of antigen and antibody, there can be hemolytic reaction, bacteriolytic reaction, and so on.

Operation method

hemolytic reaction (breakdown of red blood cells leading anemia)

Principle

The hemolytic reaction occurs only in certain bacteria (e.g., Vibrio cholerae). Application of the hemolytic reaction is an indispensable element of the complement binding reaction. Hemolysis occurs when an antigen (red blood cell) binds specifically to an antibody (hemolysin) and attracts complement, which causes the red blood cell to be lysed in the presence of the complement.

Materials and Instruments

Sheep red blood cells
Hemolysin, guinea pig serum, physiological saline.
Small test tubes Water bath

Move

1. Take 3 small test tubes and add the substances according to the table below (volume in milliliters).
2. Place the 3 test tubes in a 37℃ water bath for 15~30 minutes to observe any coagulation phenomenon.
