Cryptosporidium oocyst staining method
Cryptosporidium oocyst staining method can be divided into aureamide-phenol staining method, modified antacid staining method and aureamide-phenol staining-modified antacid staining method.
1. Auramine-phenol staining method
(1) Preparation of staining solution: 1g/L aurantium-phenol staining solution (the first solution): 0.1g of aurantium, 5.0g of carbolic acid, 100ml of distilled water; 3% hydrochloric acid alcohol (the second solution): 3 ml of hydrochloric acid, 100ml of 95% alcohol; 5 g/L potassium permanganate (the third solution): 0.5g of potassium permanganate, 100ml of distilled water.
(2) Staining procedure: add drops of the first solution to the dried fecal membrane, wash after 10-15 minutes; add drops of the second solution, wash after 1 minute; add drops of the third solution, wash after 1 minute and leave to dry; examine under a fluorescence microscope.
2. Modified antacid staining method
(1) Preparation of staining solution: carbonate red staining solution (first solution): 4g of alkaline red, 20ml of 95% alcohol, 8ml of carbonate, 100ml of distilled water; 10% sulfuric acid solution (second solution): 10ml of pure sulfuric acid, 90ml of distilled water (while stirring the sulfuric acid poured into the water); 20 g/L malachite green solution (third solution): 1ml of 20 g/L malachite green solution, 10ml of distilled water. Distilled water 10 ml.
(2) Staining steps: add drops of the first solution to the dried fecal membrane, wash after 1.5-10 minutes; add drops of the second solution, wash after 1-10 minutes; add drops of the third solution, wash after 1 minute and leave to dry; observe under the microscope.
