X-ray crystallography for protein structure determination

Summary

X-ray diffraction is an extremely important method for determining the crystal structure of proteins. Biomolecule X-ray crystallography is the science of revealing the structure and function of molecules. There is no tool with which one can directly observe the arrangement of atoms and groups inside proteins. Although electron microscopy comes close to seeing the outlines of macromolecules. But it is still limited to exposing the size, shape, symmetry and aggregation state of the molecule.
