Animal model of liver blood stasis syndrome

Summary

Liver and blood stasis syndrome animal model, refers to the animal liver qi stagnation, blood flow is not smooth, blood blockage caused by the performance of the syndrome model. At present, the method of establishing the animal model of liver and blood stasis is duck hepatitis B virus (DHBV) method.

Principle

The basic principle of duck hepatitis B virus (DHBV)-induced hepatic blood stasis is to use duck hepatitis B virus (DHBV) to induce hepatic blood stasis by repeated attacks.

Principle

The basic principle of duck hepatitis B virus (DHBV)-induced hepatic blood stasis is to use duck hepatitis B virus (DHBV) to induce hepatic blood stasis by repeated attacks.

Materials and Instruments

Laboratory animals:
① Duck (50-60 g, 1 day old)
Virus strain:
① DHBV positive serum, each 1 mL of serum contains about 3.0×10-9% DHBV.

DHBV positive serum, containing about 3.0×10 9 DHBV particles per 1 mL of serum.

Move

Duck hepatitis B virus (DHBV)-induced hepatic blood stasis was modeled as follows:
A. Ducks at 1 day of age were injected with DHBV-positive serum in the tibial vein on the same day (0.1 mL per animal).
B. On the 10th day, blood was collected from the jugular vein at a dose of 1 mL, and the serum was separated from the blood to test for DHBsAg to confirm that the infection success rate was 98%, and the uninfected animals were removed.
C. The DHBV-positive serum continued to be injected in the tibial vein at 0.1 mL per animal once a week. DHBV-positive serum was continued to be injected into the tibial vein at a dose of 0.1 mL per animal once/week.
D. The dose was increased to 0.2 mL per animal once/week starting from the 10th week.
E. Until the end of the experiment, a total of 112 days were allowed.
