Product introduction:

Nerve terminal probes are a series of cationic styryl fluorescent dyes used to track synaptic activity at neuromuscular junctions or synapses. Such dyes usually have lipophilic tails (two carbon chains) and highly hydrophilic heads with cations. The nervegreentm series of probes are dyes with a single double bond. Cationic styryl dyes function by activity dependent staining of synaptic vesicles. When the dye is co incubated with cells or tissues, the aqueous part of the dye has no fluorescence, while the lipophilic tail of the dye inserts into the cell membrane and exhibits strong fluorescence. After nerve stimulation, the dye is encapsulated in the vesicles during endocytosis. Therefore, after washing the dye attached to the cell surface, the fluorescence signal intensity indicates the number of newly formed vesicles. On the contrary, during exocytosis, the dye is released from the vesicles together with neurotransmitters, resulting in the reduction of fluorescence signal. Therefore, the change in fluorescence intensity reflects the situation of endocytosis / exocytosis or synaptic activity. The rate of fluorescence increase during endocytosis - "binding rate" and the rate of fluorescence decrease during exocytosis - "dissociation rate" vary according to the dye type. In general, dyes with longer lipophilic tails and more double bonds have higher affinity for membranes and thus higher binding rates and lower dissociation rates. This product is equivalent to FM1-43. Nervegreentm C4 is a widely used green fluorescent dye that tracks synaptic activity by staining synaptic vesicles or neuromuscular junctions at synapses. Using the red fluorescent dye nerveredtm in combination, synapses and neuromuscular junctions can be imaged in two colors independently.
