Oxygen Radical Absorbance Capacity (ORAC) is a classic method for detecting the antioxidant capacity of biomolecules in various samples. The ORAC method offers distinct advantages for evaluating antioxidant performance, including high specificity, excellent sensitivity, a broad detection range, and suitability for high-throughput screening of antioxidant activity. Consequently, it is increasingly applied in the assessment of natural product antioxidant extracts and is widely adopted by the food and functional food industries as a key evaluation standard for functional foods. The detection principle of this kit is as follows: Sodium fluorescein is used as the fluorescent probe, and the azo compound AAPH serves as the source of peroxyl radicals. The radicals degrade the fluorescent probe, causing a change in fluorescence intensity. The extent of this change reflects the degree of radical-induced damage. In the presence of antioxidants, they can inhibit the fluorescence change caused by the radicals. The degree of inhibition reflects their antioxidant capacity against the radicals.
