It is known that ELF-1 appears in the cytoplasm as a 80 KDa protein that is O -glycosylated and phosphorylated in order to be translocated into the nucleus where it can be detected as a 98 KDa protein. After dephosphorylation, the protein is degraded through the proteasome pathway. The inactive form of Elf-1 is an 80-kDa protein that lacks DNA-binding activity and is confined to the cytoplasm of the cell. Phosphorylation and O-linked glycosylation increase the molecular weight of Elf-1 to 98 kDa, the active form; 98 kDa Elf-1 binds to the promoter of the gene that codes for CD3ζ inducing its transcription. ELF1 Antibody (Ab101487) - IHC Immunohistochemical analysis of ELF1 staining in human ovarian cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with ELF1 Antibody (Ab101487) at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
ELF1 Antibody (Ab101487) - IF/ICC Immunofluorescent analysis of ELF1 staining in K562 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ELF1 Antibody (Ab101487) in 3% BSA-PBS and incubated overnight at 4 °C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.
