The purpose of fixation is to preserve the original morphology and structure of cells and tissues. Fixatives prevent autolysis of tissues and cells by endogenous lysosomal enzymes and inhibit the growth of bacteria and molds. Fixatives alter the internal structure of proteins through coagulation, formation of additive compounds, and other mechanisms, thereby inactivating enzymes. Fixative solutions are mainly classified into aldehyde fixatives, mercuric fixatives, alcoholic fixatives, oxidizing fixatives, picrate fixatives, etc. Among the commonly used ones are formalin from aldehydes and ethanol from alcohols.
FPA Fixative Solution (FPA Fluid) is mainly composed of formalin, ethanol and acetic acid. It is used for fixing general plant specimens. Fixation for 24 hours yields better results than FAA fixative and allows long‑term preservation of specimens.
Procedure (For Reference Only):
1. Fixation generally requires 24 hours.
2. Specimens can be stored long‑term after fixation.
Precautions:
1. FPA fixative is harmful to the human body. Please handle with care in a well‑ventilated environment and avoid inhalation.
2. Fixation time varies with the thickness of tissue samples. Proper tissue preparation facilitates penetration of the fixative. The suitable thickness for routine biopsy tissues is 2–4 mm, generally not exceeding 6 mm.
3. The volume of fixative should be sufficient; the volume ratio of fixative to tissue block should generally be greater than 10:1. If the volume is inadequate, the fixative may be replaced 1–3 times during fixation.
4. Temperature significantly affects fixation. Increasing temperature accelerates fixation, but the temperature should not be excessively high.
5. Fresh tissues should be fixed promptly after collection. If immediate fixation is not possible, tissues should be stored in physiological saline and submitted for testing without delay.
