Glycerol-Ethanol Mounting Medium, also known as Glycerol-Ethanol Mountant, is mainly composed of glycerol, ethanol, deionized water and other ingredients. As a water-soluble mounting medium, it is suitable for specimens that do not require complete dehydration or are prone to shrinkage and deformation after dehydration (e.g., fungi, algae, etc.), and is especially applicable for the mounting of fungi and insects. However, it has the disadvantage of easy water loss after mounting, which causes the medium to dry and shrink and form a large number of crystals. For long-term preservation, the edges of the coverslip need to be sealed with Canada balsam, neutral balsam or similar sealants. Based on the dosage of 50-100 μl of mountant per sample, 5 ml of the medium can be used for mounting at least 50 samples. This reagent is for research use only (RUO) and not intended for clinical diagnosis or any other applications.
Materials to Be Prepared by the User
Slides, coverslips, fluorescence microscope
Operating Procedures (For Reference Only)
1. Adherent Cell Samples
① Aspirate the staining solution after completion of staining.
② Add 1 drop of Glycerol-Ethanol Mounting Medium onto the slide, cover with the coverslip with adherent cells attached, ensure the cells contact the mountant, and avoid air bubbles as much as possible.
③ Observe under a microscope.
2. Tissue Sections
① Aspirate the staining solution after completion of staining.
② Add 1 drop of Glycerol-Ethanol Mounting Medium onto the tissue section, cover with a coverslip, ensure the section contacts the mountant, and avoid air bubbles.
③ Observe the tissue section under a microscope.
3. Other Samples
Operate with reference to the procedures for cell samples or tissue sections for other types of samples.
Precautions
1. All fluorescent substances are susceptible to quenching; stained samples should be stored away from light.
2. After mounting with this product, it is recommended to seal the edges of the coverslip with neutral balsam.
3. For your safety and health, wear a lab coat and disposable gloves during operation.
4. Use the reagent as soon as possible after opening to prevent adverse effects on subsequent experimental results.
