Overview

Acetyl-CoA (Acetyl-Coenzyme A) is widely present in animals, plants, microorganisms, and cultured cells. It is a crucial intermediate metabolite produced during the metabolism of energy substances in living organisms and serves as a pivotal hub in the body's energy metabolism. The three major nutrients—carbohydrates, fats, and proteins—converge via Acetyl-CoA into a common metabolic pathway: the tricarboxylic acid cycle and oxidative phosphorylation. Through this pathway, they are completely oxidized to carbon dioxide and water, releasing energy for ATP synthesis. Furthermore, Acetyl-CoA acts as a precursor for the synthesis of physiologically active substances such as fatty acids, ketone bodies, cholesterol, and their derivatives. The detection principle of this kit is that malate dehydrogenase catalyzes the conversion of malate and NAD to oxaloacetate and NADH. Citrate synthase catalyzes the conversion of Acetyl-CoA and oxaloacetate to citrate and coenzyme A. Utilizing the coupled reactions of malate dehydrogenase and citrate synthase, the amount of Acetyl-CoA is proportional to the generation rate of NADH. NADH has a characteristic absorption peak at 340 nm. By measuring the absorbance at 340 nm, the content of Acetyl-CoA can be calculated.
