This chapter is intended as a practical guide for researchers who need to modify antibodies themselves. It covers common chemical and protein hydrolysis modifications that preserve the antigen-binding function of IgG antibodies. Overly aggressive modifications are not discussed here because they result in extensive protein denaturation and functional inactivation. Radiolabeling of antibodies is also beyond the scope of this chapter.
This chapter will provide a detailed reaction flow of a typical modification, as well as an explanation of the underlying principles, but will not cover all aspects of the modification reaction.
