Experiments on the preparation of suspension-type liquid pharmaceuticals

Summary

Suspension liquid pharmaceuticals refers to the insoluble solid drugs to the particle state dispersed in the liquid dispersion medium formed by the non-homogeneous liquid pharmaceuticals. Due to the effect of gravity, the particles in the suspension will settle when left to stand. In order to make the particles settle slowly, the drug should be selected with fine particles and add the suspending agent to increase the viscosity of the dispersion medium. In addition, the addition of wetting agents and other methods can be used to increase the stability of the suspension.

Move

1. Preparation of glycerite lotion
[Prescription].
Glycerite 150 g Zinc oxide 50 g Glycerin 50 ml Sodium carboxymethyl cellulose 2.5 g Purified water Total 1000 ml
[Method]

Take glycerite, zinc oxide research fine sieve, add glycerol and appropriate amount of purified water into a paste, and take sodium carboxymethylcellulose plus purified water to dissolve, add the above paste liquid in batches, with the addition of stirring, and then add purified water to make 1000 ml, stirring, that is to obtain.

[Role and use]

Protect skin, astringent, anti-inflammatory. For skin inflammation, such as pimples, subacute dermatitis, eczema, urticaria.

[Usage and Dosage]

Shake well before use, apply externally and topically.

[Note]
(1) There are two kinds of zinc oxide: heavy and light, and it is better to choose the light one.
(2) Glycerite and zinc oxide are both hydrophilic drugs insoluble in water and can be wetted by water. Therefore, first add glycerin and a small amount of water to grind into a paste, and then mixed with aqueous sodium carboxymethylcellulose, so that the formation of a hydrated film around the particles in order to impede the polymerization of the particles, and easy to re-dispersed when shaking.
2. Preparation of compound sulfur (sulfur) lotion
[Prescription]
Zinc sulfate 30 g Sedimentation sulfur 30 g Spiritus camphoratus 250 ml Glycerin 100 ml Sodium carboxymethyl cellulose 5 g Purified water Total 1000 ml
[Method]

Take sodium carboxymethyl cellulose, add appropriate amount of purified water, stir rapidly, so that the slurry; another take the precipitation of sulfur in turn with glycerol research until fine, and the former mixture. Then take zinc sulfate dissolved in 200 ml of purified water, filtered, the filtrate slowly added to the above mixture, and then slowly add camphor spiritus, with the addition of research, and finally add purified water to 1000 ml, stir, that is obtained.

[Effects and Uses]

Protect the skin, inhibit sebum secretion, mild sterilization and astringent. Used for dry seborrhea and acne.

[Usage and Dosage]

Shake well before use and apply topically.

[Notes]
(1) Sulfur for medicinal use is divided into refined sulfur, precipitated sulfur, and sublimated sulfur due to different processing methods. Among them, the particles of precipitated sulfur are the finest, and it is easy to make fine and easily dispersed finished products, so it is better to use precipitated sulfur.
(2) Sulfur is a strong hydrophobic substance, the surface of the particles is easy to adsorb air and form air film, so it is easy to collect and float on the liquid surface, it should be moistened with glycerin and ground, so that it is easy to be mixed and suspended with other drugs.
(3) Spirit of camphor should be added into the mixture slowly with a fine stream and stirred rapidly to avoid precipitation of larger particles of camphor.
(4) Sodium carboxymethyl cellulose can increase the viscosity of the dispersing medium, and can be adsorbed around the particles to form a protective film, which makes the product stable.
(5) This product is forbidden to use soft soap, because it can generate insoluble bivalent soap with zinc sulfate.
3. Preparation of Compound Belladonna Compound
[Prescription].
Tincture of belladonna 50 ml Tincture of camphor 200 ml Tincture of orange peel 20 ml Hydroxyphenethyl ester solution (5%) 6 ml Simple syrup 120 ml Purified water Total 1000 ml
[Preparation]

Take tincture of belladonna, compound camphor tincture, orange peel tincture, hydroxybenzene ethyl ester solution (5%), mix well, slowly add this mixture into about 500 ml of purified water, with the addition of stirring, add simple syrup, and then add purified water to make 1000 ml, stir well, divided into packages, that is to obtain.

[Role and use]

Antispasmodic and analgesic. There is to relieve smooth muscle spasm and other effects. Used to relieve pain caused by smooth muscle spasm and stop diarrhea.

[Usage and Dosage]

Oral, 10 ml once, 3 times a day.

[Note]
(1) This product is a light yellow-brown suspension with a sweet, slightly bitter taste.
(2) Dry mouth may occur after taking this product, which is caused by belladonna inhibiting the secretion of glands, so more water should be drunk.
(3) Belladonna preparations should not be taken at the same time with cholinergic drugs.
(4) Belladonna is contraindicated in patients with glaucoma.
4. Quality inspection of suspension and evaluation of stabilizer effect.
(1) Determination of settling volume ratio: Pour glycerine lotion, compound sulfur lotion and compound belladonna combination into a graduated stoppered measuring cylinder, stopper it tightly, shake it vigorously for 1 minute, record the beginning height of the suspension, Ho, and place it, determine the height of the settled material, H, in accordance with the time stipulated in Table 3, and then calculate the settling volume ratio of the individual placing time according to the formula (settling volume ratio, F=H/Ho), and record it in Table 3. The settlement volume ratio is between 0 and 1. The larger the value, the more stable the suspension is.
Table 3 Settling volume ratio in 2 hours (H / Ho)
(2) re-dispersion test: the above respectively equipped with glycerite lotion, compound sulfur lotion and compound belladonna combination of the stoppered cylinder placed for a certain period of time (48 hours or 1 week later, but also according to the conditions), so that its settlement, and then the stoppered cylinder inverted flip (a reverse and a positive for a time), and will be at the bottom of the cylinder to re-dispersion of the number of times required for the turn over is recorded in Table 4. The fewer the number of turns required, the better the re-dispersion of the suspension. If there is a consistent failure to disperse, indicating agglomeration should also be recorded.

5. Preparation of triiodide combination
[Prescription]
Sulfadiazine (fine powder) 25 g Sulfadiazine (fine powder) 37.5 g Sulfadimethylpyrimidine (fine powder) 37.5 g Bulb fat 1.5 g Benzoic acid 2 g Simple syrup 200 ml Distilled water Add to 1000 ml

Caveat

1. Suspension aids should be prepared in a thick liquid of a certain concentration. Solid drugs are generally preferred to be finely ground and sieved.

2. Dispersing method of preparation of suspension, it is appropriate to use the liquid grinding method.

3. when preparing suspension by changing the nature of the solvent precipitation method, the alcoholic preparations (such as tincture, spiritus, fluidized ointment) should be added slowly into the aqueous solution with a fine stream, and stirred rapidly.

4. The vial should not be too full, should leave appropriate space for shaking before use. And should be affixed with a label printed with the words "Shake well before use" or "Shake well before serving".
